On January 31, 2007, the FDA announced that package inserts have been updated for two antiretrovirals: efavirenz and T-20. The new inserts include the following recommendations.
Efavirenz should not be coadministered with astemizole, bepridil, cisapride, midazolam, pimozide, triazolam, or ergot derivatives. Otherwise, efavirenz will compete with these drugs for CYP3A4, potentially inhibiting their metabolism and causing serious adverse events, such as cardiac arrhythmias, prolonged sedation, and depressed respiratory function.
When efavirenz and voriconazole are coadministered at standard doses, the effectiveness of voriconazole may be reduced, and the risk for efavirenz-related side effects may be increased. To avoid these problems,…
