Thymidine analogue–associated lipoatrophy is a troublesome adverse side effect of d4T and AZT. No effective treatments are currently available, other than switching off the offending drug if doing so is an option. Uridine, an exogenous pyrimidine, could potentially reverse the cellular pyrimidine deficiency induced by d4T and AZT mitochondrial toxicity.
To determine the effect of uridine on total limb fat in patients with d4T- or AZT-associated lipoatrophy, investigators randomized 20 such patients to receive uridine supplements (NucleomaxX; 36 g three times daily for 10 consecutive days per month) or placebo for 3 months.
