In the POWER studies, 11 protease mutations were found to be associated with resistance to darunavir, and the presence of 3 or more of these mutations significantly diminished response to the drug (ACC Mar 15 2006). Now, investigators have examined the prevalence and covariates of these 11 mutations in two large cohorts of HIV-infected patients.
Among 2744 PI-experienced patients in the Stanford HIV Drug Resistance Database, 24% had at least 1 of the 11 mutations. Similarly, among 1175 patients in a clinical cohort who had been treated with PI-based regimens, 30% had one or more of these mutations.
