Current guidelines recommend that antiretroviral therapy (ART) consist of at least three drugs, but could ritonavir-boosted darunavir alone be sufficient?
In the manufacturer-sponsored MONET trial, 256 patients with prolonged virologic suppression (viral loads <50 copies/mL for ≥6 months) on their current ART regimens were randomized to switch to ritonavir-boosted darunavir (800 mg of darunavir, with 100 mg of ritonavir, each once daily), either alone or with two investigator-chosen NRTIs. Treatment failure was defined as virologic failure (2 consecutive viral loads >50 copies/mL by week 48) or any discontinuation of study drugs other than NRTI changes for toxicity.
