Integrase inhibitors have become important components of many antiretroviral therapy (ART) regimens. Dolutegravir, an investigational agent in this class, has potent in vitro efficacy and can be administered once daily without boosting.
In a manufacturer-sponsored, phase II study (SPRING-1), 205 HIV-infected, treatment-naive patients were randomized to receive dolutegravir at one of three doses (10 mg, 25 mg, or 50 mg once daily) or efavirenz, each in combination with tenofovir/FTC or abacavir/3TC. The study was open label with respect to drug assignment, but blinded with respect to dolutegravir dose.
