Context

Patients with advanced prostate cancer who have homologous recombination repair (HRR) mutations, especially those with BRCA 1 or 2 alterations, have worse outcomes. The AMPLITUDE phase III study demonstrated that poly(ADP-ribose) polymerase (PARP) inhibitors improve progression-free survival (PFS) in patients with androgen pathway modulation–sensitive (APMS) metastatic prostate cancer. Now, researchers report the findings of the international, randomized TALAPRO-3 trial, in which almost 600 patients with metastatic APMS and HRR alterations received either the PARP inhibitor talazoparib or placebo.
