New antithrombotic strategies are required to reduce cardiovascular complications after percutaneous coronary intervention without increasing bleeding risk. The highly selective antagonist SCH 530348 inhibits the platelet thrombin receptor PAR-1; its use represents a novel approach to blocking thrombin-mediated platelet activation.
In a manufacturer-sponsored, multicenter study, researchers randomly assigned 1030 patients to receive oral SCH 530348 (10 mg, 20 mg, or 40 mg) or matching placebo at least 1 hour before undergoing elective angiography with intent to perform PCI. Patients who subsequently underwent PCI received a maintenance dose of SCH 530348 (0.5 mg, 1.0 mg, or 2.5 mg daily) or placebo for 60 days. All patients received aspirin …
