Long-term use of proton-pump inhibitors (PPIs) might increase risk for benign fundic-gland polyps. To examine this issue, investigators in the Netherlands prospectively evaluated 599 consecutive patients who underwent upper endoscopy. The presence or absence of fundic-gland polyps was noted in each patient; gastric mucosa and polyp biopsy specimens were collected. Survey answers about PPI use (dosage and duration) were validated using office or hospital records.
Fundic-gland polyps were noted in 75 (23%) of 322 patients who were receiving PPIs at the time of endoscopy and in 32 (12%) of 277 non–PPI users. Risk for fundic-gland polyps varied with duration of PPI use. Long-term (≥5 years) PPI use was associated with a fourfold higher risk (odd…
