In some epidemiologic studies, folic-acid supplementation has been associated with lower risk for colorectal neoplasia. Now, the Polyp Prevention Study Group has conducted a double-blind controlled trial to determine whether such supplementation might prevent recurrence of colorectal adenomas. A total of 1021 adults with recent histories of adenomas were randomized to receive daily folic acid (1 mg) or placebo and then were randomized again to receive daily aspirin (81 mg or 325 mg) or placebo. All participants underwent complete colonoscopy, with removal of any known polyps, within 3 months of enrollment. Those who completed the follow-up examinations were included in the analyses, even if they discontinued randomized supplementation.
