As a class, thiazolidinediones are associated with fluid retention and congestive heart failure. In 2007, several meta-analyses suggested that rosiglitazone (Avandia) also leads to excess risks for myocardial infarction and cardiac death (JW Gen Med May 24 2007). The manufacturer issued an interim analysis of a postmarketing cardiovascular safety study, which showed that rosiglitazone was not associated with significant excess cardiovascular deaths or hospitalizations, or for MIs (JW Gen Med Jun 12 2007); a black-box warning was added to rosiglitazone’s label (in which, evidence about risk for myocardial ischemia is termed “inconclusive”).
