The most important positive study in GI cancer is the open-label phase III ToGA trial, in which researchers evaluated the efficacy and safety of adding trastuzumab to standard chemotherapy in patients with human epidermal growth factor receptor 2 (HER2)-positive metastatic esophagogastric cancer. Patients who received trastuzumab plus chemotherapy achieved longer overall survival (OS; 13.8 months vs. 11.1 months; hazard ratio, 0.74; P= 0.0046), longer progression-free survival (PFS; 6.7 months vs. 5.5 months; HR, 0.71; P=0.0002), and higher response rates (47% vs. 35%; P=0.0017) than those who received chemotherapy alone; patients who received trastuzumab did not experience greater toxicity.
