The tyrosine kinase inhibitors sunitinib and sorafenib are used to treat patients who have renal cell carcinomas and other malignancies. Because these agents target and inhibit the vascular endothelial growth factor (VEGF) receptor, they might raise risks for bleeding and thrombosis by altering endothelial cell function.
To examine risk for arterial thromboembolic events (ATEs) in patients who received sunitinib and sorafenib, investigators performed a systematic review and meta-analysis. Ten trials comprising more than 10,000 patients (most with renal cell carcinomas) were included; most studies were phase II single-arm trials or expanded-access program studies.
