Pancreatic cancer is the fourth leading cause of cancer-related deaths, and overall survival (OS) at 5 years is a dismal 6%. Gemcitabine remains the standard therapy for metastatic disease, with a response rate of 6%, a median survival of 5 to 6 months, and a 1-year survival rate of <20%. Phase III trials adding cytotoxic or targeted chemotherapeutic agents to gemcitabine have nearly all failed to improve OS compared to gemcitabine alone.
