Autoantigens induce immunologic tolerance and have been shown to prevent type 1 diabetes in mice. In a double-blind clinical trial, Swedish investigators examined the safety and efficacy of the autoantigen glutamic acid decarboxylase (GAD) — a major autoantibody in patients with type 1 diabetes. Seventy patients (age range, 10–18 years) with recent-onset (within 18 months) type 1 diabetes, fasting C-peptide levels >0.1 nmol/L, and GAD antibodies were randomized to receive daily doses of insulin plus two injections (on days 1 and 30) of either recombinant human GAD-alum or alum alone.
