Despite the documented effectiveness of tamoxifen and raloxifene in preventing breast cancer, these selective estrogen-receptor modulators (SERMs) have not been widely used by high-risk women. Investigators examined updated findings from seven randomized trials of these older SERMs, along with results from two trials of the newer agents, lasofoxifene and arzoxifene, in women without breast cancer. The nine studies included 83,399 women followed for a median of 65 months.
SERMs were associated with a 38% reduction in incidence of breast cancer, including ductal carcinoma in situ. The estimated 10-year cumulative incidence of breast cancer was 6.3% for placebo and 4.2% for SERMs; 42 women would need to be treated to prevent one case of breast …
