The current understanding of Alzheimer disease (AD) is that the initial AD pathology develops in situ while the patient is cognitively normal. This state has been classified as predementia AD or mild cognitive impairment (MCI), a condition in which patients are usually only moderately impaired in memory, with relative preservation of other cognitive domains. MCI patients progress to AD at a yearly rate of 10% to 15% (Alzheimers Dement 2012; 8:S1). AD is a complex disease characterized by an accumulation of β-amyloid (Aβ) plaques.
