Dapsone was synthesized more than 100 years ago and is still in common use for infectious diseases, including leprosy and neutrophilic skin disorders. A rare but serious adverse effect is drug-induced hypersensitivity syndrome or DRESS (drug reaction with eosinophilia and systemic symptoms). Up to 3.6% of dapsone recipients develop this syndrome, and as many as 13% of these die as a result. A multinational team of investigators completed a genomewide association study of 872 dapsone recipients (39 with dapsone-induced DRESS and 833 controls) to identify risk factors associated with development of the hypersensitivity syndrome.
