Median survival with first-line chemotherapy for metastatic gastric cancer is less than 1 year, and second-line chemotherapy with taxanes or irinotecan confers only a modest survival benefit. Trastuzumab, which targets the human epidermal growth factor receptor 2 (HER2), is the only targeted therapy approved for gastric cancer, but it is used only in the minority of patients with HER2-positive disease.
