Targeting the B-cell receptor pathway has proven to be therapeutically effective in a variety of B-cell lymphoproliferative disorders. To test the activity of idelalisib, an oral inhibitor of PI3Kδ, in this setting, investigators conducted an industry-sponsored, phase II, open-label study of 125 patients with relapsed indolent non-Hodgkin lymphoma; (median age, 64; median number of prior therapies, 4; range, 2–12). Eligible patients — who had follicular, small lymphocytic, marginal zone, and lymphoplasmacytic lymphomas and progressive disease that was refractory to alkylating agents and rituximab — received idelalisib (150 mg orally twice daily) until disease progression or unacceptable toxicity.
