Elevated lipoprotein-associated phospholipase A2 (LpPLA2) levels are associated with plaque vulnerability and an increased risk for ischemic events. Darapladib, a selective oral inhibitor of LpPLA2, might play a role in stabilizing plaque and improving cardiovascular outcomes.
In the double-blind, manufacturer-sponsored STABILITY trial, whose top-line results were reported in 2013, investigators randomized 15,828 patients with stable CHD to receive either once-daily darapladib (160 mg) or placebo, in addition to guideline-recommended care, and followed for a median of 3.7 years.
