Although the median survival of patients with primary myelofibrosis (PMF) is 5.8 years, the survival range is quite broad, suggesting that patient characteristics as well as disease factors affect prognosis. Previous studies showed that mutations in an RNA splicing gene, SRSF2, were associated with a poorer outcome (NEJM JW Oncol Hematol Dec 4 2012). Whether other mutations, such as those in JAK2, CALR, and MPL, affect prognosis is unclear.
To examine the effect of these genetic subtypes on the clinical course, leukemic transformation, and survival of patients with PMF, investigators in Italy analyzed data from 617 PMF patients subdivided according to genotype.
