Current management of patients with advanced clear cell renal cancer involves the use of a variety of agents that target vascular endothelial growth factor (VEGF) and mammalian target of rapamycin (mTOR).
To test whether combining these agents would be more effective than monotherapy, investigators conducted a phase II trial of 331 patients (median age, 61) with metastatic clear cell renal cancer who were randomized to one of four different treatment regimens: bevacizumab alone (B), bevacizumab plus sorafenib (BS), bevacizumab plus temsirolimus (BT), and sorafenib plus temsirolimus (ST). Most patients had pure clear cell histology, and nearly three fourths had favorable or intermediate-risk disease.
