To assess whether transplant preparative regimens containing anti–T-lymphocyte globulin (ATG) would decrease the incidence of chronic graft-versus-host disease (GVHD), investigators conducted an industry-funded, prospective, randomized, open-label, phase III trial involving 155 patients with acute leukemia undergoing hematopoietic stem cell transplantation (HSCT). Patients were randomized to either ATG (10 mg/kg) daily for 3 days prior to transplantation or no ATG (control group); post-transplant GVHD prophylaxis included cyclosporine and methotrexate.
Results were as follows after 2 years of follow-up:
