Patients with Gaucher disease or obesity have an increased risk for developing multiple myeloma (MM). Because Gaucher patients have increased levels of the lysolipid LGL-1, investigators assessed the reactivity of the monoclonal immunoglobulin (Ig) paraprotein against LGL-1 and another lysolipid, lysophosphatidylcholine (LPC). Antigen-specific immunoblotting and ELISA assays were used to detect anti–LGL-1 and anti-LPC in Gaucher-associated MM and in sporadic MM cases.
The monoclonal Ig in 17 of 20 patients with Gaucher-associated MM had anti–LGL-1 reactivity, and the monoclonal Ig in 22 of 66 patients with sporadic MM had anti–LGL-1 or anti-LPC reactivity. These monoclonal Igs did not react against lipid-A, cardiolipin, or diacylglycerol. Th…
