Deutetrabenazine is a formulation of tetrabenazine that contains deuterium. The deuterium dampens CYP2D6 metabolism and increases the half-life of the drug. To evaluate safety and efficacy of deutetrabenazine for the treatment of chorea in Huntington disease, researchers conducted a double-blind study, with 90 patients randomized to deutetrabenazine or placebo. Both groups were titrated to an optimal dose during an 8-week period. This dose was maintained for 1 month, followed by a 1-week washout period. The primary outcome variable was the change from baseline maximal chorea score to the mean of the chorea score calculated at the 9- and 12-week visits. A 2.7-unit treatment difference was required for efficacy.
