Two previous phase I/II trials showed that combining dexamethasone with pomalidomide — an immunomodulatory drug approved for the treatment of refractory or relapsed multiple myeloma — had efficacy in patients with amyloid light-chain (AL) amyloidosis.
To examine the safety and impact on survival of this drug combination, investigators conducted a phase II trial involving 28 patients with AL amyloidosis previously treated with bortezomib regimens. Pomalidomide (2–4 mg daily) was given continuously in 28-day cycles; dexamethasone (20–40 mg) was given weekly.
