Proton-Pump Inhibitors and Dementia Revisited

No association between PPI use and deterioration of cognitive functions was shown in a large prospective cohort study of women.

A recent pharmacoepidemiologic analysis of a German insurance claims database suggests that proton-pump inhibitors (PPIs) are associated with an increased risk for dementia (NEJM JW Gastroenterol Apr 2016 and JAMA Neurol 2016; 73:410).

To further investigate this link, researchers conducted a nested cohort study within the large population-based, prospective U.S Nurses' Health Study II involving women middle-aged and older. They assessed medication use in nearly 14,000 participants who completed self-administered, well-validated neuropsychological testing to assess cognitive domains recognized as predictors of dementia.
