TP53 Mutations Are Prognostic in Mantle Cell Lymphoma

Mutations identified younger patients who had poor survival despite receiving intensive induction therapy.

The standard of care for younger and medically fit patients with previously untreated mantle cell lymphoma (MCL) is a regimen incorporating high-dose cytarabine and rituximab followed by consolidation therapy with autologous stem cell transplantation (ASCT).

To determine whether the presence or absence of mutations in eight genes recurrently mutated in MCL correlate with patient outcomes, investigators analyzed genetic aberrations in diagnostic bone marrow samples from 183 MCL patients in two Nordic clinical trials that used current treatment regimens (NEJM JW Oncol Hematol Nov 2008 and Blood 2008; 112:2687 and 2014; 123:2953).
