Patients with newly diagnosed multiple myeloma who are ineligible for consolidation therapy with autologous stem cell transplantation are typically treated with one of several combination regimens. To test the efficacy of combining daratumumab — an anti-CD38 monoclonal antibody approved for use in the relapsed setting — with bortezomib, melphalan, and prednisone (BMP), investigators conducted an industry-sponsored, multicenter, randomized phase III trial involving 700 patients. Patients received up to 9 cycles of BMP; daratumumab was given weekly in cycle 1, every 3 weeks in cycles 2 through 9, and then every 4 weeks until disease progression or toxicity.
