Doravirine (DOR) is an investigational once-daily nonnucleoside reverse transcriptase inhibitor (NNRTI) that retains activity against common NNRTI mutations including K103N, Y181C and G190A. It has few drug–drug interactions and is potent. Earlier studies suggested that adverse effects, including neuropsychiatric effects, are limited. In this phase 3, manufacturer-funded, randomized, controlled, double-blind noninferiority study (DRIVE-FORWARD), investigators compared DOR to ritonavir-boosted darunavir, each combined with two NRTIs, for treatment of antiretroviral-naive adults with HIV infection. In each treatment arm, 383 participants were analyzed.
