A major hallmark of Parkinson disease and related disorders is the abnormal accumulation and propagation of α-synuclein pathology in the central (CNS) and peripheral nervous systems. In cellular and preclinical models of Parkinsonism, accumulation of intracellular aggregated α-synuclein and associated pathology can be mitigated by antibodies directed to pathologic forms of α-synuclein. PRX002 is a humanized monoclonal antibody that targets the C-terminus of α-synuclein and preferentially binds its aggregated form. In a prior phase I trial, researchers demonstrated safety and tolerability of single infusions of ascending doses of PRX002.
