In a prior study, first-line chemotherapy with FOLFIRINOX was shown to be superior to gemcitabine for patients with metastatic pancreatic cancer (NEJM JW Oncol Hematol Jul 2011 and N Engl J Med 2011 May; 364:1817).
To compare these two regimens in the adjuvant setting, investigators conducted an industry-funded, international, randomized, open-label phase III trial in which 493 patients received 6 months of gemcitabine or FOLFIRINOX (infusional 5-FU, irinotecan, and oxaliplatin) after R0 or R1 resection of pancreatic adenocarcinoma. The median age of patients was 63 to 64 years, most were male (55%–58%), most had R0 resection (55%–60%), and most had stage IIB disease (73%–74%).
