Beyond VEGF and EGFR, there are limited targets for approved agents in metastatic colorectal cancer (mCRC). Immune checkpoint inhibitors benefit only the rare patient with an MSI-high tumor. HER2 amplification or overexpression is potentially targetable but is present in fewer than 5% of patients.
Investigators now report results of an industry-funded, multicenter, open-label, phase II trial (MyPathway) evaluating the combination of pertuzumab plus trastuzumab in 57 patients with chemotherapy-refractory, HER2-amplified mCRC.
