Introduction of inactivated poliovirus vaccine (IPV) into global immunization programs led to a global shortage, prompting evaluation of dose-sparing strategies. In a randomized controlled noninferiority trial in Bangladesh, investigators assessed immunogenicity of fractional-dose IPV (fIPV), measuring seroconversion and antibody titers for polioviruses types 1, 2, and 3. They randomized >1000 infants to one of four dosing schedules:
1. Full-dose IPV at 14 and 22 weeks
2. IPV at 14 weeks and fIPV at 22 weeks
3. IPV at 6 weeks and fIPV at 22 weeks
4. Three doses of fIPV (at 6, 14, and 22 weeks)
Fractional IPV (0.1 mL) was given intradermally using a HelmJect autodisable syringe; full-dose IPV (0.5 mL) was administered by needle-syringe intramuscu…
