Background and Objective

Previously, primaquine was the only drug available to kill dormant liver-stage malaria parasites and prevent relapse. This guideline summarizes tafenoquine trials and provides guidance for its use.

Key Points

Confirm normal levels of glucose-6-phosphate dehydrogenase (G6PD) before using tafenoquine.
Tafenoquine should be taken with food.
Tafenoquine should not be used in persons with hypersensitivity to 8-aminoquinolines or with a history of psychiatric disorder.
Tafenoquine is contraindicated in pregnancy and during breast-feeding, unless the infant is known to have normal G6PD levels.
The drug is an option for prophylaxis against all species of malaria in people aged 18 or above.
