Long-acting antiretroviral drugs are of intense interest for HIV treatment as well as PrEP. In HPTN 077, a placebo-controlled phase IIa trial conducted at eight international sites, researchers evaluated the safety and pharmacokinetics of a long-acting (LA) intramuscular (IM) formulation of the integrase inhibitor cabotegravir. HIV-negative subjects were randomized 3:1 to receive either of two IM regimens (three 800-mg doses every 12 weeks or five 600-mg doses for one 4-week interval, then every 8 weeks) or IM placebo. Subjects were followed for up to 76 weeks after the last injection. This prespecified analysis focused on the pharmacokinetic tail phase after the last IM dose.
