Both lenvatinib and pembrolizumab have demonstrated single-agent activity in advanced renal cancer, as has everolimus, which is FDA-approved both as a single agent and in combination with lenvatinib. In a multinational phase 3 trial, patients with previously untreated advanced renal cell carcinoma were randomly assigned to receive lenvatinib plus pembrolizumab (Len/Pem; lenvatinib administered daily; pembrolizumab intravenously every 3 weeks), lenvatinib plus everolimus (Len/Eve; both administered daily), or sunitinib (4 weeks on, 2 weeks off).
