Studies of chimeric antigen receptor T-cell (CAR-T) therapies have shown activity in relapsed/refractory multiple myeloma (R/R MM). In the latest advance, a phase 1b/2, open-label, pharma-sponsored clinical trial of the B-cell maturation antigen (BCMA)-targeting CAR-T, ciltacabtagene autoleucel, assessed the overall response rate (ORR) and toxicity in relapsed or refractory, heavily pretreated MM.
One hundred thirteen patients were enrolled; 16 did not receive CAR-T infusion due to disease progression, study withdrawal, or death. There were no failures in manufacturing of the CAR-T cell product. Ninety-seven patients (29 in phase 1b and 68 in phase 2) received the recommended phase 2 dose of 0.75 x 106 cells/kg.
