Chronic graft-versus-host disease (cGVHD) negatively impacts quality of life in many patients following allogeneic stem cell transplantation. Based on early promising results for ruxolitinib, a Janus kinase inhibitor, investigators compared ruxolitinib (10 mg twice daily) to investigator's choice of 10 different immunosuppressive second-line agents in patients with cGVHD resistant to or dependent on ongoing glucocorticoid therapy. The industry-funded multicenter randomized phase 3 trial enrolled 329 patients aged ≥12 years with moderate to severe cGVHD. Patients failing or intolerant to a control therapy at 24 weeks could crossover to ruxolitinib; 37% crossed over.
