Anti–SARS-CoV-2 monoclonal antibodies have been used for COVID-19 treatment and post-exposure prophylaxis. Now, the manufacturer-sponsored phase 3 PROVENT trial provides efficacy data on a combination of two antibodies with extended half-lives — tixagevimab plus cilgavimab — for COVID-19 pre-exposure prophylaxis. The study enrolled adults at high risk for COVID-19 progression or exposure (3.8% of whom were immunocompromised). Enrollees were excluded if they had received a COVID-19 vaccine; for those who received vaccine during the study, their data were not included after the date of vaccination. In all, 5197 participants were randomized 2:1 to receive a single intramuscular injection of tixagevimab/cilgavimab (150 mg each) or placebo.
