Plasma tau biomarkers for Alzheimer disease (AD), especially total tau (t-tau), may also be seen in peripheral tissues and may not be specific to the brain. Researchers from this study developed a novel, antibody-based, brain-derived tau (BD-tau) biomarker and tested it in five independent cohorts (n=609): two cohorts with cerebrospinal fluid (CSF) and serum biomarkers, two clinically based cohorts with plasma or serum biomarkers, and an autopsy cohort. The TauJ.5H3 antibody was designed to bind selectively to exons 4 to 5 of tau. Additional biomarkers analyzed included t-tau and neurofilament light chain (NfL). Those with a clinical non-AD diagnosis included participants with variants of frontotemporal dementia.
