Could survival in Parkinson disease (PD) depend on the causative gene, such as SNCA, PRKN, LRRK2, or GBA? To find out, investigators used data from the French Parkinson Disease Genetics Network national multicenter cohort. Subjects were genotyped for the presence of one of the four mutations. The national death register was used to document the endpoint.
Of 2037 database patients diagnosed with PD during a period of about 30 years, 889 died. Longer survival was associated with PRKN (n=100; hazard ratio, 0.41; P=0.001) and LRRK2 mutations (n=51; HR, 0.49; P=0.023) compared with those without one of the four mutations of interest. Shorter survival was associated with SNCA (n=20; HR, 9.88; P<0.001) and GBA mutations (n=173; HR, 1.33; P=0.048).
