Serogroup A meningococcal disease in Africa was virtually eliminated with the introduction of monovalent meningococcal vaccine, but disease caused by other serogroups emerged. Investigators in Mali and Gambia randomized participants in three age groups (2–10, 11–17, and 18–29 years; 51% female) to receive pentavalent ACWYX conjugate vaccine (NmCV-5) or quadrivalent ACWY conjugate vaccine (MenACWY-D). The two primary endpoints were noninferiority of immune responses as measured by serum bactericidal assay (SBA) to A, C, W, and Y antigens in NmCV-5 recipients versus MenACWY-D recipients, and to the X antigen in NmCV-5 recipients compared with the lowest response to A, C, W, and Y antigens observed in MenACWY-D recipients.
