Loss of estradiol at the menopause transition results in thermoregulatory dysfunction, manifesting as vasomotor symptoms (VMS) in many women. Fezolinetant (Veozah), a neurokinin-3–receptor antagonist that restores thermoregulatory sensitivity, recently has been approved by the U.S. FDA for managing VMS.
Three recent industry-sponsored, randomized trials provided data to support the drug's approval. Taken together, comparisons between 45-mg fezolinetant and placebo showed the following:
At 12 weeks, mean number of daily hot flashes had decreased from ≈11 at baseline to ≈7 with placebo and to 4 or 5 with fezolinetant — a significant difference of 2 or 3 episodes daily.
