Subcutaneous tirzepatide (Mounjaro), a glucagon-like peptide-1 (GLP-1) receptor agonist and glucose-dependent insulinotropic polypeptide receptor agonist, is U.S. FDA approved for use at doses as high as 15 mg weekly for treating patients with type 2 diabetes. Two large trials now have demonstrated its effectiveness for treating patients with obesity.
In a study from 2022, 2500 adults without diabetes but with obesity (body-mass index [BMI] ≥30 kg/m2 or BMI ≥27 with at least one weight-related complication) were randomized to lifestyle modification plus subcutaneous tirzepatide (at doses of 5 mg, 10 mg, or 15 mg weekly) or placebo for 72 weeks, including a 20-week dose-escalation period.
