Vicriviroc is one of two CCR5 inhibitors being developed to treat HIV infection. In this phase II study (ACTG 5211), investigators tested the safety and efficacy of vicriviroc among 118 treatment-experienced patients who had CCR5-tropic virus only and who were experiencing virologic failure on ritonavir-containing regimens. Participants were randomized to receive placebo or one of three doses of vicriviroc (5, 10, or 15 mg once daily); the assigned treatment was added to the failing regimen for the first 14 days of the study and then to an optimized background regimen. The primary endpoint was change in viral load at day 14.
Both the 10-mg and 15-mg doses of vicriviroc were superior to placebo at 14 days and at 24-week follow-up visits. The …
