Ultrashort regimens for treating latent tuberculosis infection (LTBI) caused by susceptible strains of Mycobacterium tuberculosis would be very helpful. To facilitate research in this area, investigators developed a murine model of LTBI in which mice are vaccinated with a strain of bacillus Calmette-Guérin, followed 6 weeks later by an aerosol challenge with M. tuberculosis. Six weeks after this, the mice have approximately 100,000 (4 log10) colony-forming units (CFUs) of M. tuberculosis per lung — a level that is maintained over the next 7 months. In a previous study using this model, the investigators demonstrated that rifapentine was more effective at preventing relapse of TB than isoniazid plus rifampin (JW Infect Dis Dec 23 2009).
