Multiple retrospective case-control studies have suggested a possible association between proton-pump inhibitor (PPI) use and risk for hip fractures, but these analyses were subject to potential confounding and did not prove causation. A recent study that controlled for many potential confounders showed no link between PPI use and loss of bone-mineral density (JW Gastroenterol Jan 8 2010).
Now, researchers have again investigated the possible association between hip fractures and acid-reduction therapy with PPIs or histamine-2–receptor antagonists (H2RAs). Using data from a northern California health system, they retrospectively analyzed PPI and H2RA use among 33,752 people with hip or femur fractures and 130,471 matched controls.
