The PARK11 linkage region at 2q36-37 was initially identified in a genomewide screen of sibling pairs affected with Parkinson disease in the PROGENI study (Am J Hum Genet 2002; 71:124 and 2003; 72:1053). Other researchers subsequently reported evidence suggesting that the gene for Grb10-interacting GYF protein 2 (GIGYF2) was responsible for the PARK11 linkage assignment (Am J Hum Genet 2008; 82:822). Several single amino acid changes in GIGYF2 were identified that met suggestive criteria for identifying a disease gene, including occurrence at evolutionarily conserved residues and absence in geographically matched controls.
