Certain molecular-genetic aberrations can predict a meaningful, albeit transient, tumor response to antiangiogenic therapies, especially aberrations in the antiangiogenic, cell-signaling vascular endothelial growth factor receptor (Curr Treat Options Oncol 2008; 9:1 and J Clin Oncol 2007; 25:4722). The theory of de novo tumor vasculogenesis aims to explain the transient nature of such responses and the altered vasculature seen in a subset of tumors exposed to antiangiogenic therapy. This theory is based not only on clinical and radiographic evidence, but also on basic and translational studies reporting on neural, vascular, and glioma cell signaling and other mechanisms.
